چکیده

Subtype H1N1 influenza A virus (IAV) that caused a pandemic in 2009, called A(H1N1)pdm09, has been circulating as seasonal IAV among humans, escaping from herd immunity through amino acid substitutions hemagglutinin (HA). Additionally, position 162 of HA A(H1N1)pdm09 evolved to be N-linked glycosylated by 2017. Here the effect glycosylation at was examined comparing ratio nonsynonymous diversity (πN) synonymous (πS) (πN/πS) structurally defined categories sites between 1785 (162+) and 7562 (162−) sequences were not considered 162, respectively. In 162− sequences, πN/πS for located inside 15 Å significantly smaller than one greater outside Å. 162+ corresponding values different each other. These results suggest may shield epitopes against antibodies A(H1N1)pdm09.

N-Linked Glycosylation in the Hemagglutinin of Influenza A Viruses

Since the 1918 influenza A virus (IAV) pandemic, H1N1 viruses have circulated in human populations. The hemagglutinin (HA) of IAV determines viral antigenicity and often undergoes N-linked glycosylation (NLG) at several sites. Interestingly, structural analysis of the 1918 and 2009 H1N1 pandemic viruses revealed antigenic similarities attributable to the conserved epitopes and the NLG statuses ...

Evolutionary Dynamics of N-Glycosylation Sites of Influenza Virus Hemagglutinin

The hemagglutinin protein of influenza virus bears several sites of N-linked asparagine glycosylation. The number and location of these sites varies with strain and substrain. The human H3 hemagglutinin has gained several glycosylation sites on the antigenically important globular head since its introduction to humans, presumably due to selection. Although there is abundant evidence that glycos...

Positive selection for gains of N-linked glycosylation sites in hemagglutinin during evolution of H3N2 human influenza A virus.

The number of N-linked glycosylation sites in the globular head of hemagglutinin (HA) has increased during evolution of H3N2 human influenza A virus. Here natural selection operating on the gains of N-linked glycosylation sites was examined by using the single-site analysis and the single-substitution analysis. In the single-site analysis, positive selection was not inferred at the amino acid s...

N-glycan analysis by CGE-LIF: profiling influenza A virus hemagglutinin N-glycosylation during vaccine production.

Glycoproteins, such as monoclonal antibodies as well as recombinant and viral proteins produced in mammalian cell culture play an important role in manufacturing of many biopharmaceuticals. To ensure consisting quality of the corresponding products, glycosylation profiles have to be tightly controlled, as glycosylation affects important properties of the corresponding proteins, including bioact...

Influenza virus hemagglutinin: a model for protein N-glycosylation in recombinant Escherichia coli.

BACKGROUND The hemagglutinin molecule of influenza virus is considered as an ideal model to study biological processes as well as the effect of glycosylation on the function of glycoproteins. OBJECTIVES The large subunit of the influenza virus A/New Caledonia/20/99 (H1N1) hemagglutinin (HA1) was expressed in recombinant Escherichia coli containing the glycosylation system of Campylobacter jej...
